With the backpack, drag a sprite, costume, script, or sound into it and you can take that object with you and use it in other projects.
The Make-A-Block function lets you make a block to execute a script. You can have variables, booleans, and strings for inputs in your custom block, which means you can use them in that block's scripts.

dr3w8 explained the backpack pretty well.
If you only want to know what make-a-block does, ignore this. Otherwise, what you do is, once you've ‘made’ a block and have its ‘define’ block out, you drag the blocks you want to use and attach them to it. Once you've done this, those blocks will run anywhere the block you made appears in the script.
Others could probably explain better than me me, but I hope this helps anyway.

Great explanations!
Think of making a block as making a cake. When you ‘define’ it, you are deciding the ingredients of the cake. Then you have a new ‘cake’ which is made of the ingredients.
Hopefully a simple explanation!
Hope I helped!
